Results:

AVIDIN developed and optimized several screening technologies based on different label-free methods in order to increase its capacity for in-house development programmes as well as by acting as a CRO for early drug discovery projects.

Label-free real-time cell analysis by using the xCelligence platform

A new innovative platform, the xCELLigence System (Roche Applied / ACEA Biosciences Inc.) has been purchased and the technology has been optimized for different screening projects. It monitors cellular events in real time without the incorporation of labels. The System measures electrical impedance across interdigitated micro-electrodes integrated on the bottom of tissue culture E-Plates. The impedance measurement provides quantitative information about the biological status of the cells, including cell number, viability, and morphology. A wide range of cell-based assays for both high throughput screening was optimized and several screening programme were successfuly performed.

Avidin has optimized a screening strategy based on the xCelligence system to screen cytotoxic and cytoprotective compounds (Ozsvari et al. A cell-microelectronic sensing technique for the screening of cytoprotective compounds. Int J Mol Med. 2010 Apr;25(4):525-30.) and to analyze cell migration of primary cells as well as different cell lines.

Label-free 3D Cell analysis by using holographic microscopy

A new innovative microscopic technology, HoloMonitor™ M3 (Phiab) has been purchased and the technology has been optimized for different screening projects. Phase Holographic Imaging´s HoloMonitor™ M3 is the first totally non-invasive live cell imaging microscope/cell analyzer.

Avidin is using the technology for evaluating/screening in primary cells, cancer cells and for testing immune functions, toxicity of different drug candidates.

Protein-protein/ Protein-small molecule interaction screening by using the Enspire Multimode plate reader

A new, label-free screening platform, the Enspire system (Perkin Elmer) has been purchased and the technology has been optimized for different screening projects.

Avidin is using the technology for screening small molecules for interaction with G-protein coupled receptors, fragment-based screening and for target-driven drug discovery projects.
